This film has two major points of focus: the Buddhist text, the Heart Sutra, which is part of the Ultimate Wisdom Text of Buddhism, and the travel diary of the 7th Chinese monk Xuanzang, who undertook a 10,000-mile and 16-year-long journey to India to Vulture's Peak, where the Heart Sutra is supposed to have been delivered. The Heart Sutra is a dialog between two prominent figures in Buddhism: Arya Avalokiteshvara and Arya Shariputra. This dialog is supposed to have happened in the presence of the Lord Buddha at the top of a hill called Griddhra Kut, or Vulture's Peak, in Rajgir, Bihar, India. The place still exists today. This film has interviews with the following scholars and thinkers: His Holiness the Dalai Lama, B. Alan Wallace, Barbara Du Bois, Beate Stolte-Overtheil, Bernie Glassman, Dean Brian Baker, Geshe Dorji Damdul, Geshe Ngawang Samten, Joan Halifax, Lama Zopa Rinpoche, Matthieu Ricard, Natalie Goldberg, Paul Ekman, Ravindra Panth, Robert Thurman, Tanahashi Kaz, Thupten Jinpa, and Tsultrim Allione. Following the discovery in 1995 of a set of human remains in a coffin on the coast of Patagonia in Argentina, a team of scientists set out to find out if they belonged to the long-lost grave of Catherine Roberts, the first Welsh woman to die in Patagonia shortly after she arrived with the first group of settlers in 1865. A first investigation of the DNA of the bones revealed that they belonged to a woman in her 40s of northern European origin, which matched Catherine. But to solve the mystery once and for all, they needed to find a descendant to compare the DNA. Enter Nia Olwen Ritchie, a fire fighter from present-day northern Wales to travelled to Argentina to donate her DNA and solve the mystery.

Mamadrama combines film clips, cultural commentary, interviews with Hollywood and Israeli filmmakers and footage from Schwarz's earlier films in an exploration of the image of the Jewish mother in film beginning with early silent and Yiddish films up through contemporary movies. Hollywood directors Paul Mazursky, Paul Bogart, Larry Peerce and actress Lainie Kazan reflect on their Jewish mothers. Critics Patricia Erens, J. Hoberman, Michael Medved, Amy Kronish and Sharon Rivo discuss the changing image of the Jewish mother on screen. Israeli filmmakers Avram Hefner and Zepel Yeshurun and actress Gila Almagor illustrate the uniqueness of Israeli filmic images. Mamadrama includes selections from Come Blow Your Horn, Goodbye Columbus, Next Stop Greenwich Village, Jazz Singer, Portnoy's Complaint, Where's Poppa, Torch Song Trilogy, a compilation of rare Yiddish films and recent Israeli features.
