Unhappy with their marriages, best friends, Meet, Prem and Amar, decide to cheat on their wives - but instead become the prime suspects in a murder investigation.
A murder is committed in a town in India and the Police ask a senior policeman to deal with his matter. His name is Sikanker and he begins his investigations and after doing so, he finds that he has three suspects. Three men, who are all married, but have been involved in seducing a woman named Monica. The three men are Amar, Prem, and Meet, who all claim their innocence - but in Sikander's books - they are all guilty until they prove themselves innocent. Meet, Amar and Prem have been married for the last 3 years and bored of their quirky wives. So when they meet for the first time in three years, they decide to indulge in affairs and "get some action". They meet again after a month to compare notes, but discover each one of them has had a liaison with a wily woman Monica. She blackmails her into bringing her a lot of money or else she would inform their wives of their infidelity. Masti revolves around three bachelors, Meet (Vivek Oberoi), Prem (Aftab Shivdasani) and Amar (Ritesh Deshmukh). Their lives are good until they get married, and their lives turn to hell.
Meet marries Anchal (Amrita Rao), who is the daughter of a rich jeweler. Anchal is obsessively possessive about her husband and makes him wear traditional Indian clothes. Anchal calls Meet every half an hour and wants to know everything he is doing at all times. Meet feels suffocated due to this.
Prem marries Geeta (Tara Sharma) who is overly religious and thus their sex life suffers.
Amar marries Bindiya (Genelia D'Souza) who is dominating and lives with her mother (Archana Puran Singh). Bindiya starves Amar of sex and makes him exercise all the time, serving him nothing but bland and boiled food for nourishment. Amar is a renowned dentist, but Bindiya gives him only enough money to take an auto-rickshaw to and from work.
Dr. Suresh Kapadia (Satish Shah) runs his sexologist practice in the same office complex as Amar and is always seen with beautiful nurses by his side. Suresh tells Amar to hire a good-looking nurse for his own office, which will be good for business.The friends get together one night and try to think of a way to get away from their wives for some time and relive their fun bachelor days. The men set their sights on other women and decide that they will try for one month and then get together again to exchange tales of their affairs.
Sheetal (Divya Palat) is Meet's employee at the jewelry store. He takes Sheetal out for lunch but is interrupted when Anchal walks into the same restaurant. Meet abandons Sheetal and keeps Anchal away from her.Amar decides to treat his patient Neha Saxena (Rakhi Sawant), who has the hots for Amar, and he had been avoiding giving her an appointment. Amar goes out on a date with Neha, but she turns out to be a dude and he runs away.Prem spots a sexy customer in his bank, and offers to deliver her passbook to her home at night. Prem anticipates an amorous encounter but turns out the customer had many kids.
One day Monica walks into Meet's store to exchange a necklace, to which he agrees. That night Meet goes to Monica's apartment with an intention to seduce her, but she asks him to slow things down.
The guys eventually realize they have all been seeing the same girl, Monica (Lara Dutta). She blackmails them about revealing their affair to their wives for Rs.10 Lakhs each.Monica says that she had heard the guys talk about having an affair a month ago and made her plan. She then bought jewelry at Meet's store, had a dental appointment with Amar and opened a bank account with Prem.
The guys try various tricks to get the money from their wives. Amar decides to rob his own Mother-in-law when she is alone in the house. Meet generates a fake letter from his uncle who needs Rs 10 Lakhs as emergency help for his daughter's dowry. Prem's wife entrusts Prem with Rs 12 Lakh to deposit in bank on behalf of her charity. The charity money was donations that Geeta had collected over several months.
The terrified men arrive at Monica's car with all the money, only to find her dead. So that they are not accused of Monica's death, they try to hide her body. They are interrupted by police officer Sikander (Ajay Devgan) who begins to suspect, and then starts following them when he finds Monica's dead body near the area where he found the boys.Meanwhile, the wives who were aloof till now, suddenly turn all sexy and turned on and give a lot of attention to their husbands. But the husbands are not able to consummate their love as they are interrupted by visions of Monica.
Sikander finds out that Monica had dealings with all 3, she had purchased jewelry at Meet's store, opened an account via Prem and went for a dentist appointment to Amar's clinic.Thereafter, the trio goes to Monica's house to retrieve the photos that link them to Monica. Inspector Sikander traces the three who hide at Monica's veranda.
Next morning, a mysterious man (Shahbaz Khan) finds them and reveals that he killed Monica and demands ransom to cover the crime. When Monica was found dead Amar had kept all the 30 Lakhs in his mother-in-law's safe.Now since the money is required again, he plans to take his mother-in-law and wife for a movie, so that Meet and Prem can steal the money. But that night, real robbers enter the house. Amar & his wife & mom-in-law return early from the movie. Mother-in-law engages Prem & Meet in hand-to-hand combat on the roof (as they try to enter the house), while Amar helps the real robbers run away with the cash (thinking of them to be Prem & Meet).
The next day, Amar's wife calls the police & again Sikander arrives. He brings the real robbers in, who confirm that Amar was the one who handed the case over to them, as mistook them for his friends. Amar's wife is outraged at this accusation and defends him.
The trio, guilt-ridden goes to their respective wives to apologize. But the very next day, the killer traces them. Sikander reaches the same spot & the killer forces Meet to drive to escape. during the chase, Amar Prem ends up jostling with the killer for the gun and accidentally shoots him.
Eventually the police catch up with them and they end up in jail. After their wives show up, it is revealed that it was just a prank played on them and reveals Monica is alive and the killer turns to be a police officer named Vinayak Bapat.The men then apologize to their wives and promise to never do "Masti" again.
