A comedy centered around three people who each have a lookalike of a lookalike, all with the same name.
Millionaire Ashok and his friend Kumar are admitted to a psychiatric hospital. When Ashok's Mamaji and Dr. Khan mix medicine into the guys' water, they both start to behave like dogs. Later, their Mamaji's look-alike Johnny is also admitted to their hospital.—[email protected]
Ashok (Saif Ali Khan) and Kumar (Ritesh Deshmukh) are best friends, whereas Shanaya (Tamannaah Bhatia) and Mishti (Bipasha Basu) are their love interests. Ashok and Kumar live in London, where Ashok aspires to be a stand-up comedian. Kumar is Ashok's manager, and Mishti manages his estate portfolio. Shanaya was the only person who laughed at Ashok's jokes, when everybody else stood up and left his show.Shanaya works for a TV Channel and hosts a reality show called "Kaun Banega Millionaire". In her show, a random person from the streets is made a millionaire for a week.
Ashok's father Mr Singhania (Akash Khurana) is a multi-millionaire but has been in a coma for the last six years, while his wicked Mamaji Kunwar Amar Nath Singh (KANS Mama) (Ram Kapoor) teams up with an evil doctor, Dr. Khan (Nawab Shah) to get rid of Ashok and take over all his property. Khan is willing help Mamaji as Mamaji has been finding his illegal experiments for many years.
Mamaji can't kill Ashok or his father, since the property will go to charity, hence Ashok has to put into coma like his father or declared mentally insane for Mamaji to get the estate. Khan gives Mamaji a Mind-Altering Drug (MAD) that he has developed from DNA of many dog species. The MAD when delivered into a person's body, alters the person's genes, and changes the Chromosome structure for 24 hours, making them behave like a dog.
During a board meeting Mamaji mixes a medicine in Ashok and Kumar's drink and they both start behaving like dogs for 24 hours. Mamaji tells the board that Ashok & Kumar get fits occasionally and the doctors have not been able to treat it.
They are taken to a mental asylum for treatment by Dr. Shivani (Esha Gupta). Dr Khan had arranged for a ward boy inside the asylum who would give them the dog serum every morning and that way Ashok and Kumar would never regain their sanity.Things go wrong, when the ward boy fails to administer the serum and Ashok & Kumar recover from their dog behavior. Shivani was interviewing them when the ward boy serves them water, and the duo again turns into dogs. Shivani checks the CCTV footage and figures out that the serum in the water is turning Ashok & Kumar into dogs and vows to release them.
But fate takes a twist as two lookalikes of Ashok and Kumar are being treated in the same hospital's 'B' ward (which is a prison for the criminally insane). They work for Bijlani (Chunkey Pandey), a cocaine smuggler. The look alike used to run a Parantha stand for Bijlani in South Hall. Bijlani has asked Ashok to safeguard a whole box of cocaine (labelled as flour) till narcotics raids were over. But Kumar, mixes the cocaine in his Paranthas & serves it to their customers. Bijlani gives them so many electric shocks that they are rendered mentally insane.Ashok 2 has a crush on Shivani, while Kumar 2 likes Shanaya on her TV show.
Ashok 2 & Kumar 2 escape the fences of their ward as Ashok 2 wanted to meet Shivani.As luck would have it the duplicates are accidentally released instead of the true ones. Shanaya and Mishti, who come to pick up their boyfriends from the asylum, take away the duplicates instead, without realizing anything. Ashok & Kumar (2) think that Shanaya is taking them to a mansion as part of her show "Kaun Banega Millionaire".Meanwhile the warden of ward B Y M Raj, captures the originals and takes them back to ward B.
Shanaya and Mishti take them to Ashok's mansion. Ashok & Kumar (2) are taken to meet Mr Singhania, where (when alone) they tinker with Singhania's medical instruments, which brings him out of a coma. But then they put the instruments back in their original configuration and Singhania again reverts into a coma.Mamaji understands the whole mix-up and plans to use the duplicate Ashok to become the owner of the whole business empire.Now the true Ashok and Kumar also come to know of Mamaji's evil plan but are stuck in the asylum. They are offered help by one of the ward boy Cyrus (Darshan Jariwala), who takes them to a secret ward 'C' to meet 'Johnny' who is a look alike of Mamaji but has a dangerous habit of attacking people who sneeze in front of him.
Ashok and Kumar plan revenge against Mamaji with the help of Johnny but get caught by the warden Y. M. Raj (Satish Shah) before they can escape from the asylum. Y. M. Raj prepares to punish them but accidentally sneezes in front of them. Angered by this, Johnny beats him black and blue, thus helping the three to escape.
On the day of the board meeting Ashok & Kumar (1) get paired with Mamaji (1) and fight in the board meeting to wrest control of the company. Dr Khan sends them out to sort it out. Meanwhile Ashok & Kumar (2) get paired with Mamaji (2) and go into the board meeting fighting to hand over control of the company to each other. Ashok & Kumar (1) realize that their Mamaji is not Jhonny, but the real deal (he didn't attack a waiter who sneezed). The true Ashok and Kumar were about to expose Mamaji, when Dr. Khan triggers the fire alarm. All the three mental patients escape the building. Now desperate Ashok and Kumar bump into a dance bar owner Balbir the triplicate of Mamaji, while Mamaji also arranges another set of Ashok and Kumar with the help of plastic surgery.
Mamaji takes the triplicate Ashok & Kumar to 'House of Commons' where the fake Ashok is to hand over all the business to him in the presence of Prince Charles but the real Ashok & Kumar arrive along with the triplicate of Mamaji. To add to the confusion the three mental patients also storm in to help the real Ashok & Kumar creating panic among the eyewitnesses on seeing so many lookalikes together.Movie closes with Ashok's father coming out of his coma and recognizing the true Ashok and getting Mamaji arrested for his deeds.
